In this issue, Chen etal.1 show that genetic knockout of Piezo2 in SOX2+ medulloblastoma cells decreased local tissue stiffness, increased drug delivery across the blood-tumor barrier, and improved survival, uncovering a mechanosensitivity-dependent mechanism of action in a mouse model of pediatric brain tumors.
