Abstract

Circular RNAs (circRNAs) can be used as therapeutic targets for various human diseases through competitive endogenous RNA (ceRNA). Earlier research in our laboratory has revealed that circRNAs could serve as ceRNA to sponge microRNAs (miRNAs) and regulate the innate immune response of blunt snout bream (Megalobrama amblycephala) after Aeromonas hydrophila infection, but the specific regulation is still uncertain. In this research, by means of overexpression/RNAi, qPCR, and luciferase assay etc., we discovered a circRNA termed circARHGEF15, which was upregulated following A. hydrophila challenge and positively regulated antibacterial immunity in the host. Moreover, we observed that miR-214 can restore the promotion of inflammatory factors due to circARHGEF15 overexpression. Further, our data indicated that circARHGEF15 may function as a ceRNA of miR-214 in the immune reaction generated by A. hydrophila, reduce the inhibition of miR-214 on hepcidin, and promote the expression of downstream proinflammatory factors such as il-1β, il-6, il-8, and tnf-α. In summary, the results of this study suggest that M. amblycephala circARHGEF15 acts a significant function in the modulation of antibacterial response to A. hydrophila infection, and lays the foundation for rapid diagnosis, effective control, and treatment of fish diseases from the perspective of circRNAs in the future.
