Abstract

Exercise Exercising provides multiple benefits to the body by increasing strength, reducing disease, managing weight, and improving brain health. Using a metabolomics approach, Reddy et al. show that succinate is secreted in mice and humans during exercise by a pH-gated mechanism involving monocarboxylate transporter 1 (MCT1). The succinate that is released during exercise signals through the succinate receptor 1 (SUCNR1) to regulate the paracrine response and affect innervation and extracellular matrix remodeling in muscle. Further, SUCNR1 signaling antagonizes muscle inflammation. This work shows that paracrine regulation by the succinate-SUCNR1 pathway in muscle is important for muscle strength and innervation, as well as for restoring insulin sensitivity.
