Abstract
The following study demonstrates the effective sensing ability of nano-platform mediated microwave digestion in single cancer cells and cancer stem cells analysis using MALDI-MS. We synthesized gold nanoparticles (10–17nm) and ornamented them with biomolecules from FBS (80–120nm), for effective internalization and surface enrichment of gold nanoparticles on the cancer cells. The biological mimicking of the NP surface with biomolecules, led to its successful interaction with the cancer cells. These particles showed excellent biocompatibility. A combination of the nanoparticle mediated microwave digestion in enhancing the detection sensitivity of MALDI-MS was demonstrated on 1000, 100, 10 and finally 1 cancer cell concentration. Microwave digestion in the presence of nanoparticles enhanced the sensing ability upto 60% of MALDI-MS signals, leading to detection of molecules from a single cancer cell. The results suggest the future use of this method for the sensitive detection of single cancer cells in clinical analytes too. With this bio-mimicked nanoplatform based sensing technique, we have succeeded in obtaining MALDI MS signals from single cells of human breast origin (breast normal, cancer and cancer stem cells).
