Abstract

Bisphenol A (BPA), an endocrine disrupting compound, is widely generated in plastic products. It plays as a mixed agonist‐antagonist with respect to steroid hormones such as estrogens. Recent studies have suggested that BPA promotes adipose tissue and oxidative stress by molecular mechanisms including up/down regulation of adipogenesis and antioxidant defense systems. The aim of this study was to evaluate the effect of BPA on adipogenesis and oxidative stress during the differentiation of 3T3‐ L1 preadipocytes. We also demonstrate that effect of a combination of BPA plus beta‐carotene on lipid accumulation and ROS production in 3T3‐L1 cells. Our results show that BPA promotes the lipid accumulation and ROS production in 3T3‐L1 cells. Especially, BPA significantly increase ROS production (~40%) in 3T3‐L1 cells compared to control cells by up‐regulating gene expression of pro‐oxidant enzyme. Nevertheless, BPA in combination with beta‐carotene was markedly inhibited lipid accumulation and ROS production compared with control groups that treated with BPA alone. These results indicate that beta‐carotene consider inhibitor that acceleration of adipocyte differentiation by BPA
