DOI: 10.1002/sscp.202100062 The cover picture shows filter paper which is converted into a sample preparation unit for high-end bottom-up protein analysis. The trypsin covalently bound to cellulose digests proteins enabling shotgun and targeted proteomics experiments from dried matrix spots. These paper-based smart proteolysis samplers allow instant sample preparation starting at the moment of sampling with potential continuation during postal transport. The smart proteolysis sampler shows good stability (no activity loss for at least 4 months when refrigerated) and the proteolytic activity of bound trypsin is up to 50–60% of trypsin acting in solution
