Abstract

Vascular endothelial growth factors (VEGF) regulate hematological tumors progression. This study was aimed to assess the relationship between two VEGFA polymorphisms, C−2578A and G+405C, and the risk of non-Hodgkin's lymphoma (NHL). Blood samples were collected from a total of 246 subjects. The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique was used for genotyping. A novel in silico analysis was performed to predict the potential functions of the SNPs. CA genotype of C−2578A polymorphism was associated with NHL. We found that codominant, dominant and over-dominant inheritance models of this SNP were significantly associated with this hematological disorder. No significant link found between G+405C polymorphism and NHL risk. However, the presence of the mutant alleles of VEGFA G+405C and C−2578A polymorphisms could provide new binding sites for nuclear transcription factors. In our study, the C−2578A polymorphism conferred an increased risk of NHL. The mutant allele of this variant might affect VEGFA transcriptional activity.
