Abstract

IntroductionRheumatoid Arthritis (RA) is a relatively common inflammatory autoimmune disease. SPOCK1 is involved in extracellular matrix regulation and is implicated in RA. Publicly-available Wellcome Trust Case Control Consortium (WTCCC) RA dataset was used to select candidate RA associated variants within SPOCK1 gene for further analysis. MethodsSNPs within SPOCK1 gene, which were significantly associated or trending towards significance with RA in the WTCCC data, were investigated and rs2881668 was genotyped in New Zealand, Oxford, UK and Dutch sample sets consisting of 2693 cases with RA and 1844 controls using TaqMan® SNP genotyping assay technology. ResultsThe association analysis of SPOCK1 rs2881668 in this study (P = 0.586, OR = 1.040) failed to replicate the previously reported association of rs2881668 with RA in WTCCC data (P = 0.001, OR = 1.251). ConclusionThe findings of our study did not support the association of SPOCK1 rs2881668 with RA. Thus, our results do not support the role of SPOCK1 in RA susceptibility in the Caucasian population.
