Abstract
Penicillin considered the most familiar β-lactam antibiotic that causing allergic reactions with an incidence rate of 0.7-10% of all population. Excessive release of IL-4and IFN-γ is thought to be important in development and regulation of atopic and other allergic disorders.To estimate the possible role of IL-4RαQ576Rgenes polymorphism in development of immune reaction against penicillin,and study the effect of IL-4 and IFN-γ in regulating of allergic reactions.Measurement ofserum levels of IL-4,IFN-γ and total IgE done by using enzyme linkedd immunosorbentt assaya (ELISAa) technique. IL-4RαQ576Rgene polymorphisms were genotyped by using polymerasee chainn reactions restrictions fragments lengths polymorphisms (PCRr-RFLPp).Comparing with ccontrol subjectss,there was a significant higher levels of IL-4,IFN-γ and IgE in patients group,they were (348.53 pg/ml) versus (284.72pg/ml) and (194.33pg/ml) versus (159.82pg/ml) and (357.28pg/ml) versus (124.88pg/ml) respectively with P=0.001. The IL-4RαQ576Ralleles were significantly higher in penicillin allergic individual compared with apparently healthy control subjects. Results suggested that IL-4and IFN-γ cytokines play a crucial role in modulating of B-lactam allergy. IL-4RαQ576Rgene polymorphismsmight involvein development of penicillin hypersensitivity.
