Abstract

ADAM19 is encoded on chromosome 5q32‐q33, which has been reported to be linked with the development of asthma. Aim of study is to investigate the association of polymorphisms on ADAM19 with the risk and the phenotypes of asthma. We found 22 SNPs on ADAM19 by direct sequencing. Among these polymorphisms, 5 SNPs were selected for genotyping in Korean considering the location of SNPs, MAF and LD. In the association analysis, we could not found any association between the polymorphisms and the risk of asthma. However,+56656T>C,+95719C>T and +97188T>C showed significant association with the totalIgE level in serum of asthmatics, especially in that of atopic asthmatics (P=0.001–0.048). Asthmatics having rare allele on those SNPs showed significantly higher levels of total IgE than those having common alleles. Asthmatics having haplotype 1, which is composed by the combination of major alleles on the SNPs, showed significantly lower levels of total IgE than those having other haplotypes (p=0.002). Overall, ADAM19 may be associated with IgE level in asthma, genotypes and haplotypes of ADAM19 could be used for the new genetic markers for the hyper IgE syndrome.
