Abstract

Objective To explore the association of a single nucleotide polymorphism (SNP) rs821597 in gene disrupted-in-schizophrenia-1 (DISC1) with white matter integrity in healthy people. Methods Sixty healthy persons were genotyped at the SNP rs821597 in DISC1 and were divided into two groups (risk allele carriers and non-risk allele carriers).Diffusion tensor magnetic resonance images of the whole brains were acquired with a single-shot echo planar imaging (EPI) sequence aligned to the straight axial plane. After preprocessed with DTI-studio and SPM5 software, the fractional anisotropy (FA) images of the two groups underwent two-sample t-test with voxelbased analysis in SPM5 software. Results Risk allele carriers showed reduced white matter FA in the right caudate nucleus (MNI:14,0,16; cluster=138 voxels) and left caudate nucleus (MNI:-14,0,16; cluster=23 voxels) compared with non-risk allele carriers (P<0.05,FDR multiple comparisons). Conclusion The SNP rs821597 in gene DISC1 may be associated with white matter integrity in healthy people. Key words: Schizophrenia; Genes; Diffusion magnetic resonance imaging
