Abstract
Purpose: Asporin (ASPN) is a small leucine rich proteoglycan, which has elevated expression during intervertebral disc (IVD) degeneration and osteoarthritis in the knee. This gene is shown to be significantly associated with lumbar disc degeneration (LDD) in Chinese and Japanese population. In vitro, ASPN inhibits chondrogenesis by downregulation of TGF-β signaling, however it’s in vivo consequence and the underlying molecular mechanism for degeneration is not clear. In this study, we sought to provide insights to the role and contribution of of ASPN in LDD by generating mouse models overexpressing the human ASPN gene within the IVD.
