Abstract

The application of high-pressure liquid chromatography to quantitative analysis of ascorbic acid in biological samples is described. Experimental conditions were selected that might be representative of those met under a broad range of research and clinical trials. The technique is seen to provide values of ascorbic acid in various samples of guinea pig tissues that are in agreement with an existing analytical method. The short time required for analysis and the use of experimental apparatus that is found with increasing frequency in clinical and research laboratories make this approach attractive. The decay rates of ascorbic acid at different values of temperature and pH are evaluated. Endogenous levels of ascorbic acid in several tissues of fasted and nonfasted guinea pigs are reported.
