Abstract

The HbpA protein in Corynebacterium diphtheriae is crucial for iron acquisition from the hemoglobin-haptoglobin (Hb-Hp) complex. Lyman et al. (e00196-21) show that the C-terminal domain of HbpA is essential for cellular localization, hemin iron acquisition, and optimal hemoprotein binding. The C-terminal domain is also required for the formation of large, secreted complexes of HbpA that are involved in iron uptake from Hb-Hp. HbpA lacks homology to known hemin transport proteins and has exceptional affinity for hemoglobin. The multiple functions associated with this unusual C-terminal domain expand our understanding of the mechanism of iron acquisition by bacterial pathogens.
