Abstract

The intracellular membrane domain (IMD) is a metabolically active membrane compartment of mycobacteria associated with cell growth. The IMD proteome is not yet fully established. Rokicki et al. (e00419-21) took advantage of the Mycobacterial Systems Resource, a database that includes fluorescence microscopy of 1,153 proteins highly conserved among mycobacteria, to complement and cross-validate proteomic analysis of the IMD. They confirmed the subcellular localization of 20 previously identified IMD proteins while discovering three new ones, demonstrating the utility of this novel strategy in overcoming some of the limitations of conventional proteomics.
