Abstract

Objective To explore the application of high-resolution melting method with unlabeled probe for genotyping mitochondrial single nucleotide polymorphisms (mtSNPs). Methods we applied high-resolution melting of unlabeled probe to genotype mtSNPs of individuals and confirmed the results by direct se-quencing and restrict fragment length polymorphism (RFLP) methods. Results It shows that our genotyping results are consistent by comparing HRMA methods with direct sequencing and RFLP methods. Conclusion The study snggests that HRMA method could be a higher accuracy and a useful tool for mtSNPs genotyping. Key words: Mitochondrial DNA; Mitochondrial single nucleotide polymorphism (mtSNP) ; High-resolution melting analysis
