Abstract

Direct analysis in real time (DART)-orbitrap mass spectrometry (MS) combined with multivariate data analysis was applied to perform quality control for Yuanhu Zhitong Tablet (YZT). The ionization gas temperature of DART and extraction solvent were optimized to obtain high-quality MS spectra. Based on DART-MS data, the quantification of tetrahydropalmatine was conducted. Moreover, principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA) were utilized to investigate the differences of samples from five manufactories. The samples were classified into three groups, although the content of tetrahydropalmatine as a commonly used analytical marker showed no significant differences among the samples. Eleven ions were selected as analytical markers via multivariate and univariate data analysis, and the intensities of these ions in different samples were investigated. The results demonstrated the complementary function of these ions to conventional quality control component tetrahydropalmatine. This study exhibited the promising potential of DART-MS in quality control of YHZ and could provide us a new option in the rapid quality assessment of traditional Chinese herbal medicine (TCHM) and traditional Chinese medicine preparation (TCMP).
