Abstract
目的 探讨反义结缔组织生长因子(connective tissue grouth factro,CTGF)对兔耳增生性瘢痕的抑制作用.方法 选择20只大耳白兔建立增生性瘢痕动物模型,随机分成5组,对照组为A组,注射转化生长因子β1(TGF-β1)为B组,注射CTGF为C组,先后注射TGF-β1和CTGF为D组,注射反义CTGF为E组,每个治疗组又分为治疗7、14、20 d 3组;通过逆转录-聚合酶链式反应(RT-PCR)方法检测增生性瘢痕中CTGF mRNA的表达,免疫组织化学检测CTGF蛋白表达,HE、Masson染色观察切片内成纤维细胞数密度,计算机辅助图像分析测算切片内胶原纤维面密度.结果 E组中CTGF mRNA、蛋白表达、成纤维细胞数密度和胶原纤维面密度均降低,与A、B组比较,差异有统计学意义(P＜0.05).结论 反义CTGF可以抑制兔耳增生性瘢痕的增殖过程,使瘢痕组织纤维化程度明显减轻。
