Abstract
Objective To discuss the ethanol extracts from honeysuckle-schizonepeta spike (HSS) against influenza virus in MDCK cell and content changes of the active ingredients in HSS with different compatibilitity ratios were studied in order to screen out the best compatibility ratio. Methods The viability of influenza virus infected MDCK cell mixed with HSS was detected by MTT assay using ribavirin as a positive control drug, and the antiviral effect was evaluated. Meanwhile, the contents of chlorogenic acid, luteoloside and luteolin in HSS were determined by reversed-phase high performance liquid chromatography. Results Therapeutic index (TI) of HSS with different compatibilities (1∶0, 0∶1, 1∶1, 2∶1, 3∶1 and 4∶1) against influenza virus were 34.83, 32.92, 23.26, 87.06, 37.08 and 24.71 respectively, while the TI of ribavirin was 35.90. The virus inhibition rate of HSS with the 2∶1 group was (94.54±2.25) %, which was significantly better than ribavirin group[ (64.77±7.89) %] when the drug concentrations were both 2.0 mg/mL (t=7.25, P<0.01) . Conclusions HSS with different compatibilities all exert anti-influenza virus effect, and HSS with the compatibility ratio of 2∶1 inhibits influenza virus replication best.
