Abstract

Objective To investigate the anti-inflammatory roles of heine oxygenase-1 (HO-1)induced by ambroxol in gastric aspiration lung injury. Methods Forty Sprague-Dawley rats were randomly divided into four groups:control group,injury group,ambroxol group and znppix group. The expression of HO-1 was examined by Western blot. Leucocyte and neutrophil leucocyte in bronchoalveolar iavage fluid were counted. Tumor necrosis factor-α (TNF-α), interleukin-1ββ (IL-1β) and IL-10 in bronchoalveolar lavage fluid were examined by enzyme linked immunosorbent assay. The ultrastructure of the pulmonary alveoli and its capillaries were studied by transmissional electromicroscopy. Results Compared with control group and injury group, the gray value of HO-1 in ambroxol group increased (all P <0.05).Compared with injury group, white blood cell and neutrophil leucocyte counts,TNF-α, IL-1β and IL-10 in bronchoalveolar lavage fluid of ambroxol group decreased ( all P < 0.05 ). Compared with ambroxol group,the above indexes in znppix group increased (all P <0.05). Transmissional electromicroscopy showed that the pathological changes of ambroxol group were milder than those of injury group and znppix group. Conclusions Ambroxol may induc.e the expression of HO-1 and has anti-inflammatory effects by restraining the releasing of TNF-α, IL-1β, IL-10 in gastric aspiration lung injury.
