Abstract

SummaryThe historical and molecular derivatives of thiazide and sulphonamide diuretics from 4-sulphonamido benzoic acid are briefly discussed. In a search for new antihypertensive agents many novel 3-sulphamoyl-4-chlorobenzamides in the indoline and iso-indoline series were synthesized. These compounds were screened in a battery of pharmacological tests and, in particular, their antihypertensive and diuretic activities were investigated in experimental hypertensive rats. The research programme resulted in the selection of indapamide as a compound which produced a significant and prolonged antihypertensive action with only a slight and transient diuresis.
