Abstract
Glycoconjugates prepared by diazotizing β-( p-aminophenyl)ethylamine derivatives of oligosaccharides to the hemp seed protein, edestin, are effective immunogens. Rabbits immunized with the conjugates produce antibodies that bind radiolabeled oligosaccharides. Specificity of binding was determined by comparing inhibitory activities of structurally related oligosaccharides in radioimmunoassay. All of three rabbits immunized with a mannotetraose (Manα1-3Manα1-2Manα1-2Man) conjugate, two of five rabbits immunized with a lacto- N-fucopentaose III (Galβ1-4 [Fucα1-3]GlcN Acβ1-3Galβ1-4Glc) conjugate, and four of five rabbits immunized with a lacto- N-tetraose (Galβ1-3GlcNAcβ1-3Galβ1-4Glc) conjugate developed useful antibodies (i.e., antibodies with K a ⪖ 10 4 liters mol −1 for the corresponding oligosaccharide). These antibodies can be used to detect specific sugar sequences in complex carbohydrates.
