Abstract

Objective To detect the expression of cyclooxygenase-2 (COX-2),microvascular density (MVD),and apoptosis of bladder cancer cells in the SD rats and explore their relationship with pathologic grade.Methods Fifty SD rats were divided randomly into 5 groups,including blank group,N-methyl-N-nitrosourea (MNU) group,cinobufagin (Cino) group,adriamycin (ADM) group and bacillus calmette-guerin (BCG) group.After perfusion for one week,bladder tissues were taken out for hematoxylin and eosin (HE) staining and detection of related indicators.Results Immunohistochemical results showed that COX-2 expression,in lower-grade groups,had no significant difference among different groups.MVD expression and apoptosis of cancer cells in Cino group and ADM group showed no significant difference,but higher than in BCG group.In the higher-grade group,COX-2 and MVD expression in Cino group (2.500 ± 0.535 and 11.750 ± 1.581 respectively) was lower than the rest groups,while apoptosis rate (18.500 ± 0.535) was higher than other groups.Conclusion Cinobufagin can effectively antagonize bladder cancer and suppress the growth of bladder cancer with high grade in SD rats.
