Abstract

Background The present study was designed to determine the eff ects of cyclooxygenase-2 (COX-2) inhibitor-nimesulide on platelet activating factor (PAF) and arachidonic acid (AA)-induced platelet aggregation during myocardial infarction (MI) and ischaemia in rabbits.Methods Rabbits were divided into three groups; group I served as control and contains normal rabbits that received saline, group II rabbits received saline before induction of MI, and group III rabbits received nimesulide (25 mg/kg) before MI induction. Myocardial infarction was induced by isoproterenol (ISP) (65 mg/kg) and confi rmed by biochemical, electrophysiological and histopathological methods. Platelet aggregation was induced with arachidonic acid (AA) and platelet activating factor (PAF), and monitored by using light transmission aggregometery.Results When platelets were challenged with two diff erent aggregating agents, nimesulide-treated infarcted rabbits showed resistance to platelet aggregation while non-treated infarcted rabbits were found prone to aggregation with both aggregating agents (P >0.05).Conclusion These results suggest platelet inhibitory eff ects of nimesulide during experimental ischaemia and infarction in rabbits.
