Abstract

A novel magnetic super-hydrophilic zwitterionic hydrogel (MagG@PCBAA) was prepared via a cross-linking reaction. The MagG@PCBAA hydrogel displayed high sensitivity (0.5 fmol/μL), superior selectivity (molar ratio BSA:HRP = 10,000:1), satisfactory recovery rate (119.1 ± 1.9%), stable reusability (10 times) and high load capacity (300 mg/g). Furthermore, the MagG@PCBAA hydrogel also exhibited an excellent capability for specific capture of glycopeptides from serum of normal control and Alzheimer's disease patient. 147 glycopeptides allocated to 72 glycoproteins were captured from serum of normal control and 187 glycopeptides allocated to 84 glycoproteins were enriched from serum of Alzheimer's disease patient analyzed by Nano-LC-MS/MS. Based on gene ontology analysis, 36 primitive glycoproteins captured from the serum of Alzheimer's disease patient were significantly involved in various of neurodegenerative disease-related events, including glycoprotein binding, antigen binding and serine-type endopeptidase activity, acute-phase response, lipid transport and cholesterol metabolic process.
