Abstract

AIM: To study the anti-HIV-1 activities of Luffaculin 1 purified from Luffa acutangula. METHODS: The cytotoxicity of Luffaculin 1 was assessed by MTT assay. The anti-HIV-1 activities were determined by inhibition of syncytia formation and inhibition of p24 antigen production in HIV-1 acutely or chronically infected cells. RESULTS: The Luffaculin 1 inhibited HIV-1IIIB induced syncytium formation with EC50 values of 0.025 μmol·L 1. The therapeutic index was 76. Luf-faculin 1 also reduced p24 antigen expression in HIV-1IIIB acutely infected C8166 and in clinically isolated strain HIV-1KM018 infected PBMC, with EC50 values of 0.033μmol·L 1 and 0.207 μmol·L 1, respectively. Luffaculin 1 showed no effects on the HIV-1IIIB replication in chronically infected H9 cells. CONCLUSION: Luffaculin 1 is a novel ribosome-inactivating protein with anti-HIV-1 activity.
