Abstract

Objective To construct pulsed-field gel electrophoresis (PFGE) library typing of Yersinia pestis in Yunnan Province,and to provide a basis for control of the disease through originating and typing the disease.Methods Twenty-nine strains isolated from Rattus flavipectus plague focus,Apodemus chevrieriEothenomys miletus plague focus and Yulong plague focus in Yunnan Province were involved in this study.Two kinds of endonucleases of Asc Ⅰ and Fse Ⅰ were used in PFGE analysis,and clustering patterns were draw using BioNumerics 6.6 software.Results The 29 strains of Yersinia pestis can be divided into 20 genotypes by PFGE.These genotypes were grouped into three major clusters,namely Rattus flavipectus gene cluster,Apodemus chevrieri-Eothenomys miletus gene cluster and Yulong gene cluster.Rattus flavipectus gene cluster could be further divided into 3 sub-clusters of Dehong,Wenshan and Lincang.Conclusions There are regional aggregations of molecular typing of Yersinia pestis in Yunnan Province by PFGE; and the result of PFGE genotyping is well related to that of ecological typing; Yulong plague is an independent PFGE pattern.
