Abstract

A microfluorometric method is described for quantification of Gal-1-PO 4 and galactose in whole blood spotted on chromatography paper which makes frequent monitoring easier for the patient with galactosemia on dietary treatment. The method is sensitive enough to measure Gal-1-PO 4 levels below 3 mg/dl, and, because of incorporation of a volume correction factor, it is more accurate than other methods which rely on estimates of the volume of blood spotted on paper. These advantages, as well as the added quantification of blood galactose, should make this method applicable not only to clinical studies but also to research designed to investigate possible relationships between Gal-1-PO 4 concentration and CNS involvement.
