Abstract

A very sensitive method has been developed to measure nanogram (ng) quantities of morphine in biological fluids. The morphine is extracted from biological fluids and the extract is completely dried in a vacuum evaporator. The extracted morphine is oxidized to pseudomorphine under mildly alkaline conditions. The fluorescence intensity of pseudomorphine is then determined in a filter fluorometer. The method has been used over a wide range of concentrations, and amounts as little as 10 ng can be accurately determined. This method has been used to determine plasma and cerebrospinal fluid levels of morphine in dogs after small i.v. doses of morphine sulfate. It has also been applied in the study of morphine transport by chloroid plexus in vitro.
