Abstract
目的 介绍彗星试验(Comet assay)方法,并运用其分析人胚肺二倍体成纤维细胞(2BS)及外周血淋巴细胞(Lymphocytes)DNA损伤水平与修复能力.方法 将2BS或淋巴细胞悬液与1%低熔点琼脂糖(LMA)等体积混合,铺于0.5%正常熔点琼脂糖(NMA)胶面上,细胞经充分裂解、电泳、PI染色后,用Leica图象分析系统每样本分析100个细胞彗距和彗尾面积.结果 多数年轻与老龄2BS细胞、年轻与老龄人群淋巴细胞均无明显彗尾.28PD 2BS经H2O2损伤后彗尾明显,且彗距与彗尾面积随H2O2浓度升高而增大.批间(n=6)重复性实验显示,H2O2处理后细胞彗距s=1.7~3.5(μm)、CV=1.6%~2.8%,彗尾面积百分率s=1.5%~2.3%、CV=1.8%~2.5%.结论 年轻与老龄细胞DNA单链断裂水平均较低.H2O2可明显损伤DNA,其损伤水平随H2O2浓度升高而加大,彗星试验是评价DNA损伤水平与修复能力的较好方法。
