Abstract

For clinical diagnosis of colon tumors, breast tumors, lung cancer, ovarian carcinoma and cystadeno carcinoma, Carcinomaembryonic antigen (CEA) is a very important tumor marker. In the present study, an electrochemical immunosensor for determination of CEA biomarker was suggested. The core-shell nanofibers were prepared by electrospinning at the surface of the electrode directly and then the electrospun nanofibers were decorated with gold nanoparticles (GNPs) and multi wall carbon nanotubes (MWCNTs). Finally, CEA antibody was immobilized on the electrode surface. The steps of the process were characterized using cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) in [Fe (CN)6]3−/4− solution as a probe. The results are demonstrated that Honey nanofibers (HNF) decorated with GNPs and MWCNTs are very permeable to probe. It is a novel approach for the preparation of label-free electrochemical immunosensor. Under optimal conditions, the unique immunosensor modified with HNF exhibited high sensitivity toward CEA biomarker in the low concentration range 0.4–125 ng mL−1, with the detection limit of 0.09 ng mL−1 (S/N = 3). The results indicated that the small changes in CEA concentration can be sensed with the suggested immunosensor. Also, the immunosensor has potential in the clinical screening of cancer biomarkers.
