Abstract

Objective To study the expression of nuclear factor (NF)-κB and hypoxia inducible factor (HIF) -1 α and histopathological changes of the brain in a rabbit model after blocking the left subclavian artery and the left subclavian artery plus the left common carotid artery.Methods Twenty-four healthy adult Japanese white rabbits,regardless of sex,were randomly divided into three groups (8 rabbits per group),blocking the left subclavian artery group,blocking the left subclavian artery plus the left common carotid artery group and control group.An experimental rabbit model was set up by using thoractomy.The cerebral infarction was observed by TTC staining,histopathological changes were examined by hematoxylin and eosin (HE) staining,and the expression of NF-κB and HIF-1α was detected by using immunohistochemistry at 8th h after blockage.Results There was no sign of cerebral ischemia injury,and the expression of NF-κB and HIF-1α was lower than that of control group after blocking the left subclavian artery.However,there were obviously histopathological changes for ischemia and hypoxia,the expression levels of NF-κB and HIF-1α were higher than those in control group (P <0.05 ) after blocking the left subclavian artery plus the left common carotid artery,the results of the positive unit were (43.9538 ± 4.9238 ) and (25.0838 ±3.3532),and the results of the positive rate were (0.4468 ±0.0717)% and (0.4643 ± 0.0647) % respectively.Conclusion It has no side effects by blocking the left subclavian artery,however,it has severe damage on brain after blocking the left subclavian artery plus the left common carotid artery.It suggests that it is feasible for only covering the left subclavian artery in the clinic and it is essential for bypass surgery to supply brain blood before covering the left common carotid artery.
