Abstract

目的 探讨不同剂量FK506对神经端侧吻合功能恢复的影响.方法 30只Wistar大鼠取右侧腓总神经切断后,近端结扎并反转缝于邻近肌肉上,远端与胫神经外膜开窗处行端侧缝合.左侧为正常对照.术后分为5组,每组6只.于术后当天开始,每组于动物右小腿外侧肌肉内分别注射生理盐水(对照组)和生理盐水稀释的FK506(实验组),剂量分别为1mg/kg-1·d-1(实验组1)、2mg/kg-1·d-1(实验组2)、4mg/kg-1·d-1(实验组3)、8mg/kg-1·d-1(实验组4),连续2周.于术后3个月检测术肢双侧腓总神经、胫前肌电生理、组织学检查和胫前肌肌湿重的测定.结果实验各组全部电生理检测动作电位恢复率、肌肉单次收缩力恢复率、强直收缩力恢复率均显著高于对照组,再生神经纤维数目、肌湿重、肌纤维截面积均显著优于对照组.实验组2、3、4组之间差异无显著性意义,但均高于实验组1.结论 FK506对神经端侧吻合功能恢复具有促进作用.2mg/kg-1·d-1为比较合适的使用剂量。
