Abstract

A method is described for detecting PSTV in bulked potato leaf samples. Nucleic acids from 0.1g from each of 100 potato leaves are inoculated to tomato seedlings that are checked for viroid by PAGE of their nucleic acids after 35 days above 25°C. Viroid strains previously isolated from three tuber-bearingSolanum species were infective to tomato in sap diluted X1000 and were detected by PAGE after 28 days at about 22°C. Simulated tests showed that viroid from one infected potato leaflet in 100 was readily detected. Furthermore, diluting nucleic acid extracts from these 100-leaflet samples X10 or X100, or adding to the extracts nucleic acid from a further 20g of healthy tissue and diluting X10, did not prevent transmission to tomato. The practical use of the test is discussed, and lowered viroid concentration in potato sources grown in cool conditions considered.
