Abstract

Abstract Dual biocatalytic synthesis of D-3-hydroxybutyrate, a precursor of biodegradable polyester, poly-(3-hydoroxybutyrate), from CO2 and acetone via acetoacetate as an intermediate with enzyme extract including acetone carboxylase (AC) and 3-hydroxybutyrate dehydrogenase obtained from Rhodobacter capsulatus SB1003 cultured in acetone-bicarbonate medium in the presence of ATP and NADH is accomplished. The catalytic activity of AC was enhanced in the presence of ammonium chloride, and the conversion yield of acetone to D-3-hydroxybutyrate by the dual biocatalytic system was estimated to be up to 95% after 24 h incubation.
