Abstract

This study evaluated the ameliorative potential of hydroalcoholic extract of Trichosanthes dioica root (TDA) against arsenic (As)-induced toxicity in rat brain. TDA (5 and 10 mg kg−1) was administered orally to Wistar albino rats for 20 consecutive days before oral administration of sodium arsenite (10 mg kg−1) for 8 days. Brain biochemical parameters including lipid peroxidation, reduced and oxidized glutathione, glutathione-S-transferase, glutathione peroxidase, glutathione reductase, superoxide dismutase, catalase, and DNA fragmentation were determined. Pretreatment with TDA significantly and dose-dependently modulated all the antioxidative parameters and reduced DNA fragmentation in As-exposed rat brain. Data indicate that T. dioica root produced remarkable ameliorative effects against As-induced brain toxicity in rats mediated by modulation of oxidative stress.
