Abstract

1. 1. In primary cultures of porcine granulosa cells incubation with isoproterenol (10 −7–10 −5M) produced a dose-dependent increase in 3β-HSD mRNA, 3β-HSD content and progesterone production which ranged from 1.5- to 5-fold. 2. 2. These effects were completely blocked by alprenolol. Terbutaline (10 −6M) increased 3β-HSD mRNA, 3β-HSD content and progesterone production (1.5- to 3-fold), an effect which could be prevented by alprenolol. 3. 3. Treatment with dobutamine (10 −6M) produced no significant change in 3β-HSD mRNA accumulation. 4. 4. The results suggest that β-adrenergic agonists have the capacity to regulate transcription and translation of 3β-HSD mRNA in granulosa cells and this effect is mediated by the β 2 adrenergic receptor.
