Abstract

Extensive attention has been attracted to blocking histone deacetylases (HDACs) due to its significant over-expressing in various types of tumors. Herein, we presented two AIEgen-based water-soluble fluorescent probes, TPAsaha and TPEsaha, capable of monitoring the expression of a single HDAC isoform (HDAC6) in living cells. These two probes hardly emitted in water, and showed bright fluorescence towards HDAC6 with a very fast response time and very low interference. The fluorescence enhancement aroused from the restriction of intramolecular motion upon strong interaction with HDAC6, as proved by molecular dynamics simulations. The unique signal response mechanism makes TPAsaha and TPEsaha highly attractive in early cancer diagnosis, treatment monitoring, and prognosis assessment.
