Abstract

A mesoporous molecular SBA-15 was synthesized using Pluronics P123 (PEO20PPO70PEO20) as a structure-directing agent and TEOS as the silica source under acidic condition, and the structure of SBA-15 was determined by X-ray powder diffractometer and N2adsorption desorption. As it has been reported that siliceous SBA-15 adsorbents can have a very high affinity and capacity for proteins, the recombinant human basic fibroblast growth factor (rhbFGF) which is a kind of unstable protein was adsorpted by SBA-15, then the properties of SBA-15/bFGF complex were studied. The results had shown that the adsorption rate of rhbFGF by SBA - 15 reached 69.31% in pH 7.4 phosphate buffer at 4 °C. After 14 days, the released rate of rhbFGF from SBA-15 was 41.40% and 43.51% respectively in pH 10 phosphate buffer at 4 °C and 25 °C. Furthermore, the proliferation activity on NIH3T3 cells of the released rhbFGF still can be detected by MTT assay. Therefore, SBA-15 is possible to be used as a kind of material for bFGF adsorption and releasion.
