Abstract

Heating of a dilute adrenocorticotropin (ACTH) solution in 0.1 M NaOH has been found to cause a loss of both adrenal-stimulating and lipolytic activities. However, the same treatment not only potentiated but also prolonged the intrinsic melanocyte-stimulating potency of ACTH. Extensive physical and chemical studies of the NaOH-treated ACTH have been carried out. It was found that a limited cleavage of the second bond in the hormone occurred and that a number of amino acid residues within the molecule have been racemized. Possible explanations for the potentiation of melanocyte-stimulating activity in ACTH have been discussed.
