Abstract

SK&K 12185 [2-(p-aminophenyl)-2-phenylethylamine] inhibits in vitro corticogenesis by normal rat and guinea pig adrenals stimulated with ACTH, indicating a direct inhibitory effect on the adrenal. The acute and chronic administration of SK&K 12185 prevents the increase in peripheral plasma corticosterone levels of cold stressed rats. The peripheral plasma hydrocrotisone levels of ACTH treated guinea fails to rise following the oral administration of SK&K 12185. Manifestations of adrenal insufficiency are evident by the inability of rats to excrete an acute water load and to survive a cold stress. A natriuresis is evident in the sodium deficient rat which revieved a sodium and water load concomitantly with SK&K 12185. SK&K 12185 is goitrogenic, anethetic or estrogenic.
