Abstract

Many studies have shown that Na-K-ATPase has important role in the secretion of aqueous humor, and clinically epinephrine, and timolol are used widely to change the aqueous humor dynamics. In many tissues, norepinephrine and cAMP can either stimulate or inhibit Na-KATPase activity indirectly. Here we studied the activity change of rabbit ciliary body Na-K-ATPase after incubating with epineplirine, timolol, or cAMP in tissue homogenate. Tissue cAMP level and intraocular pressure under the influence of timolol were measured also. The results showed that epinephrine, and cAMP all had no effect on Na-K-ATPase of the rabbit iris ciliary body even at very high concentration. But timolol strongly inhibited Na-K-ATPase activity over 50% and increased tissue cAMP level 2-4 fold at a concentration over 1 mM. It also decreased the intraocular pressure 1.3-2.6 mmHg when applied topically as a 10% suspension. From these results, we thought that on rabbit ciliary body adenyl cyclase did not have any influence on NaK-ATPase and the β-receptor-adenyl cyclase pathway may have no important role in aqueous humor dynamics.
