Abstract

1. 1. The presence in rat-liver mitochondria of two inorganic pyrophosphatase activities, characterized by pH optima and Mg 2+ requirement, is reported 2. 2. The effect of substrate concentration on enzymic activity was studied; certain apparent kinetic parameters are presented. 3. 3. AMP, ADP, ATP and deoxyAMP were found to inhibit mitochondrial inorganic pyrophosphatase activity markedly and to reverse partially F- inhibition of this same activity. 4. 4. Other nucleotides and related compounds tested were found to be either completely ineffective or only very slightly active as inhibitors of this system. 5. 5. AMP inhibition of inorganic pyrophosphatase activity in aged rat-liver mitochondria was found to be competitive with respect to pyrophosphate. 6. 6. The possible significance of these findings relative to pyrophosphate-adenine nucleotide phosphotransferase activity is discussed.
