Abstract

Formaldehyde in the fish body has been occured naturally through the work of trimethylamine-NOxide<br />demethylase (TMAO)ase enzyme that breaked TMAO into formaldehyde and dimethylamine.<br />Formaldehyde fish has been occured during storage. This reserch aimed to determine of TMAOase activity<br />and its role in the formation of dimethylamine and formaldehyde lizard fish (Saurida tumbil) stored at<br />chilling temperature. Observations were made every 2 days for 16 days of storage. Analysis was performed<br />among other content of formaldehyde, dimethylamine, and specific activity of TMAOase enzyme. Result of<br />formaldehyde test increased during storage of day 0 was 0.22±0.00 ppm to 7.45±0.46 ppm on day 16. The<br />results of dimethylamine test increased during storage from 0,93±0,50 ppm at day 0 to 27.26±1.8 ppm on<br />day 16. The results of specific activity TMAOase lizardfish activity increased during storage that was 1.15 U/<br />mg on day 0, to 16.47 U/mg on day 16.<br /><br />

BAHAN DAN METODE Bahan dan Alat

Bahan utama yang digunakan adalah ikan beloso (Saurida tumbil) yang telah diidentifikasi oleh LIPI dengan bobot ±150200 gram/ekor yang diperoleh dari Tempat Pelelangan Ikan Cituis Tangerang, Banten. Sampel ikan beloso diambil secara acak sebanyak 3 ekor untuk 3 kali ulangan yang kemudian dilakukan pengamatan. Bahan kimia yang digunakan untuk analisis bahan terdiri dari tris base (Merck), HCl (Merck), asam asetat (Merck), NaCl (Merck), TCA (Merck), FeCl2 (Merck), substrat TMAO (Sigma-Aldrich), cystein (Merck), asam askorbat (Merck), DMA (Merck), bovin serum albumin (Sigma-Aldrich), Coomassie Brilliant Blue (Applichem), etanol (Merck), asam fosfat (Merck), kertas saring Whatman, pereaksi Nash (15 g amonium asetat (Merck), 0,3 mL asam asetat (Merck) dan 0,2 mL asetil aseton (Merck)), copper ammonia (25 g amonium asetat (Merck), 0,2 cooper sulfat (Merck), 25 mL NaOH 40% (Teknis), 20 mL amonia (Merck) dan akuades. Alat yang digunakan terdiri dari sentrifuge (J2-21 Beckman), spectro UVVis RS Spectrophotometer (UV-2500), mikro pipet (Thermo Scientific), pH meter (Thermo Electron), waterbath (SWBR17), vorteks (VM-300) dan alat-alat gelas (Pyrex)
