Abstract

The aim of this study was activation of cross-linked cellulase in supercritical carbon dioxide. Therefore at first, the process optimization for the preparation of cross-linked enzyme aggregates from enzyme cellulase using glutaraldehyde, was done. The precipitation was made with several precipitation reagents: acetone, ammonium sulphate, ethanol, methanol, propanol, tetrahydrofuran and 2-propanol. The initial enzyme concentration, bovine serum albumin and glutaraldehyde concentrations were optimized to obtain as high activities of the immobilized enzyme preparation as possible. Cellulase in free and immobilized form was characterized by determining enzymatic activity and aggregation yield was determined for immobilized cellulase. Cellulase cross-linked enzyme aggregates treatment with supercritical carbon dioxide for the purpose of its activation was studied, as well. Additionally, the stability and reusability of cross-linked enzyme aggregates were tested after multiple consecutive cycles. In this paper, some relevant advantages of using supercritical carbon dioxide against atmospheric pressure for cross-linked enzyme aggregates activation are presented.
