Abstract

Gene Delivery Poration methods for introducing genes into cells must achieve sufficient throughput to deliver clinically useful numbers of transfected cells. Belling et al. have enhanced the mechanical poration of cells in microcapillaries by mounting them on ultrasonic transducers. Studies with fluorescently labeled DNA were used to image delivery into Jurkat cells. Ultrasound amplitude and fluid flow rates that maintained cell viability were adjusted to maximize expression of enhanced green fluorescent protein (eGFP) plasmids. These flow rates could process 200,000 cells per minute. In peripheral blood mononuclear cells and CD34+ hematopoietic stem and progenitor cells, eGFP expression was maintained for 3 days with 80% viability. Proc. Natl. Acad. Sci.
