Abstract

This project focuses on the development of a DNA‐aptamer based biosensor, which is referred to as an aptasensor, that would aid in the detection of mycotoxins in foodstuffs. We developed aptamer based biosensor for label‐free detection of Aflatoxin B1 (AFLAB1) through the immobilization of aptamers on gold quartz crystal surface (AT‐cut/8 MHz). We used TSM, this method is sensitive to mass adsorbed at the sensor surface, measured by changes of resonant frequency, fs, and to the viscoelastic contribution, measured by motional resistance, Rm. Addition of AFLAB1 to a sensor surface with immobilised thiolate DNA aptamers resulted in subtle decrease of fs and small increase of Rm. We therefore used polyclonal antibodies anti AFLAB1 (Ab anti AFLAB1) to amplify the signal of the transducer.
