Abstract
The synthesis and chemistry of 2-(p-acetaminophenoxy)-tetrahydropyran, a prodrug of acetaminophen, are described. The utilization of a prodrug approach to the formulation of chewable acetaminophen tablets is discussed. Although the compound studied has many desired properties for use in chewable tablets, its aqueous solubility is very low so the dissolution rate may be rate determining in the release of acetaminophen.
