Abstract

Background & Objectives: Limb ischemic preconditioning provides an amenable and noninvasive approach for remote organ protection against ischemia-reperfusion injury[1]. Methods like adopting an inflatable tourniquet or a rubber band strapped around the preconditioned body to establish the limb ischemia preconditioning model is subjective, while methods using ultrasonic Doppler, laser Doppler perfusion imaging, and magnetic resonance (MR) are objective, but time-consuming and have technical restriction limitation[2]. The aim of this study was to explore a simple and reliable method to establish hindlimb ischemia preconditioning model in rats. Materials & Methods: Twenty 8–10 weeks male SD rats were randomized to two groups (group A and group B, n=10 in each group). In group A, hindlimb of the rat was strained by a tring gradually until the femoral artery blood flow was totally blocked under detection by Ultrasonic Doppler. At this moment, the tension value was record. The 99% reference range of the tension value was calculated and its upper limit was taken as F. In group B, whether the femoral artery blood flow was blocked by the string around the root of the hindlimb at the tension value F was verified by Laser Doppler. Results: In group A, the average tension value need to block the blood flow of 10 rats is 2.40±0.36 N, and the 99% reference range was [1.5, 3.3]. So the F value is 3.3 N. Under the Laser Doppler, the femoral artery blood flow in all rats of group B was completely blocked with the tension value of 3.3 N added on the string around the root of the hindlimb, which was indicated by the red arrow in figure 1.Conclusion: Applying 3.3 N tension on the two tails of a string rounding the rat hindlimb could simply and effectively block the blood flow in 8–10 weeks male SD rats, so this method could be used to establish ischemia preconditioning model on hindlimb in rat.
