Abstract

Background: QYYYG is a Chinese herbal formula. The previous study found that QYYYG can improve subclinical myocardial damage of hypertension. Mitochondrial dysfunction plays a key role in promoting cardiac hypertrophy. The emphasis of this study lies in detecting mitochondrial function and further illuminating the multi-target effect of the QYYYG. Methods: Based on transcriptome data, perform bioinformatics analysis. WKY rats as a normal control group, SHR were randomly divided into 3 groups: Model group (SHR), Valsartan group and QYYY group. Intragastric administration, once a day for 8 weeks. The mitochondrial function was detected by mtDNA. The general view and degree of fibrosis were observed by HE and Masson staining. The WGA staining was used to examine the cell size. Results: Transcriptome analysis showed that mitochondrial dysfunction was an essential mechanism of hypertensive myocardial remodeling in mice. Compared with the WKY group, SHR group HE and Masson staining showed characteristic pathological lesions including cardiomyocytes becoming thicker and apparent interstitial fibrosis. The QYYY group showed a remarkable improvement in cardiac hypertrophy and degree of fibrosis. The WGA staining was also consistent with HE staining. The QYYY group mtDNA copy number was significantly increased. Conclusions: Overall, QYYYG might improve cardiac hypertrophy by rescuing mitochondrial function. The therapeutic effects of QYYYG on cardiac hypertrophy were consistent with those suggested by transcriptome analysis. Transcriptome analysis as those described here may contribute to the holistic understanding and pharmacological evaluation of herbal remedies.
